Amtolmetin guacil is a prodrug of tolmetin and is indicated to treat pain, inflammation and fever and has been used in osteoarthritis, rheumatoid arthritis, minor rheumatic disorders, and postoperative pain. Amtolmetin guacil is a prodrug of tolmetin and exhibits anti-inflammatory, analgesic and antipyretic activity. Nonsteroidal anti-inflammatory drugs (NSAIDs) mechanism of action appears to be due to inhibition of prostaglandin synthesis in body tissues by inhibition of cyclooxygenase, an enzyme that catalyzes the formation of pro-inflammatory eicosanoid precursors from arachidonic acid. However, the exact anti-inflammatory, analgesic, and antipyretic mechanisms of action of amtolmetin guacil have not been completely clarified.
