Brigatinib is a tyrosine kinase inhibitor and cytochrome P450 3A inducer. Its indication is to treat anaplastic lymphoma kinase positive, metastatic non-small cell lung cancer (ALK+ NSCLC) that has spread to other parts of the body in people who have already been treated with crizotinib (Xalkori®) and have not improved or have worsened. Brigatinib acts as a tyrosine kinase inhibitor with activity against multiple kinases including ALK, ROS1, and insulin-like growth factor 1 (IGF-1R), and against epidermal growth factor receptor (EGFR) deletions and point mutations. It acts by inhibiting ALK phosphorylation, the activation of downstream signaling proteins, and the proliferation of cell lines expressing fusion proteins echinoderm microtubule-associated protein-like 4 (EML4)-ALK and nucleophosmin (NPM)-ALK.
