Clofazimine is an antimycobacterial used for the treatment of lepromatous leprosy. Clofazimine inhibits mycobacterial growth and preferentially binds to mycobacterial DNA, leading to disruption of the cell cycle and death of the bacterium. Clofazimine also exerts anti-inflammatory activity primarily through inhibition of T lymphocyte activation and proliferation. Interaction of clofazimine with membrane phospholipids results in the generation of antimicrobial lysophospholipids, which promote membrane dysfunction and interference with potassium ion uptake.
