Dolasetron is indicated for the treatment and prevention of nausea and vomiting postsurgical and from chemotherapy. Dolasetron mesylate, a 5-HT3 receptor antagonist, is a selective serotonin inhibitor. Antivomiting and anti-nausea effects are postulated to stem from blockade of 5-HT3 receptors located on the nerve terminals of the vagus nerve in the periphery and centrally in the chemoreceptor trigger zone of the brain. This drug has little or no affinity for dopaminergic or other serotonin receptors.
