Doxylamine antagonizes many of the effects of endogenous histamine by blocking H-1 receptors. The depressant effects are probably related to a high affinity to H-1 receptors in the brain. Many of the H-1 antagonists also tend to inhibit responses to acetylcholine that are mediated by muscarinic receptors.
