Lurasidone is indicated for treatment of schizophrenia and is a partial agonist at serotonin 5-HT 1A receptors, and antagonist at dopamine D 2 and serotonin 5-HT 2A receptors. Lurasidone is an oral antipsychotic for the treatment of schizophrenia and is a partial agonist at serotonin 5-HT1A receptors, and antagonist at dopamine D2 and serotonin 5-HT2A receptors. Lurasidone has a high affinity for D2 receptors (Ki value of 1.68 nM) and for 5-HT2A receptors (Ki value of 0.5 nM); it has moderate affinity for serotonin 5-HT1A receptors (Ki value of 6.4 nM). Antagonism of serotonin receptors can improve negative symptoms of psychoses and reduce the extrapyramidal side effects associated with typical antipsychotics.
