Maprotiline is a tetracyclic antidepressant indicated for the treatment of depression. Maprotiline inhibits presynaptic uptake of catecholamines, thereby increasing their concentration at the synaptic clefts of the brain. Maprotiline acts as an antagonist at presynaptic α2-adrenergic inhibitory auto-receptors and hetero-receptors, which increases central noradrenergic and serotonergic activity. The sedative effects of maprotiline are due to the inhibition of histamine H1 receptors.
