Phenylbutazone is indicated for use in the treatment of backache, ankylosing spondylitis, rheumatoid arthritis, and reactive arthritis. The mechanism of phenylbutazone has not been clearly established, but it is believed to bind to and inactivate prostaglandin H synthase and prostacyclin synthase through peroxide (H2O2) mediated deactivation. The reduced production of prostaglandin leads to reduced inflammation of the surrounding tissues.
