Protriptyline HCl belongs to the class of tricyclic antidepressant drugs and its mechanism of actions against depression is not clear. Tricyclic antidepressants inhibit the uptake of serotonin and norepinephrine but their antidepressant action may relate to changes in receptor sensitivity. They also affect histamine H1 receptors, α1-adrenergic receptors and muscarinic receptors, which accounts for their sedative, blood pressure lowering, and anticholinergic effects (e.g. blurred vision, dry mouth, constipation, urinary retention).
