Pyrimethamine is a folic acid antagonist used as an antimalarial or in combination with a sulfonamide to treat toxoplasmosis infections. Pyrimethamine is an antiparasitic compound and folic acid antagonist that is highly selective against plasmodia, the parasitic protozoan causing malaria, and Toxoplasma gondii, the cause of toxoplasmosis. It inhibits the dihydrofolate reductase of plasmodia and thereby blocks the biosynthesis of purine and pyrimidines, which are essential for DNA synthesis and cell multiplication. Pyrimethamine is a slow-acting blood schizontocide, is possibly active against pre-erythrocytic forms of the malaria parasite, and inhibits sporozoite development in the mosquito vector.
