Topotecan is an anti-neoplastic agent that inhibits topoisomerase I from causing reversible single strand breaks in deoxyribonucleic acid (DNA) by attaching to the topoisomerase I-DNA complex. The drug exerts cytotoxic effects when it forms a ternary complex with topoisomerase I and DNA that interacts with replication enzymes, causing double stranded DNA damage during DNA synthesis.
