Xevinapant (formerly known as AT-406, DEBIO-1143, SM-406) is an orally available mimetic of the natural second mitochondrial-derived activator of caspases (Smac) and an inhibitor of apoptosis proteins (IAPs), with potential immunomodulating, apoptotic-inducing, chemoradiosensitizing, and antineoplastic activities (1, 3). Xevinapant inhibits cancer cell growth in various human cancer cell lines, and it is under investigation for the treatment of unresectable, locally advanced head and neck squamous cell carcinoma (LA-HNSCC).
